D-Serine and Serine Racemase*

ABSTRACT:

D-serine mainly distributed in the mammalian forebrain, hippocampus and striatum, which as neurotransmitters from glial cells, target at NMDA receptors, and cause central nervous system signal transduction. A new neurotransmitter of D-serine is found playing an important role in synaptic plasticity, learning and memory. Serine racemase (SR) is a pyridoxal 5'-phosphate(PLP)-dependent enzyme that catalyzes the interconversion of Land D-serine. Serine racemase may be a new target for the neurological and psychiatric diseases. In this review, the recent advances in D-serine and serine racemase with the aspects of structure, function, mechanism and regulation will be discussed.

摘 要:

D-丝氨酸主要存在于哺乳动物的前脑,海马区和纹状体,作为胶质细胞释放的神经递质,作用于NMDA (N-methyl-D-aspartate receptor)受体,引起中枢神经系统中信号的传导。在突触可塑性,学习和记忆等方面起重要作用。丝氨酸消旋酶是一种磷酸吡多醛依赖酶,在ATP和Mg2+的辅助下,通过消旋作用将L型丝氨酸转变为D型丝氨酸。对于神经及精神性疾病,丝氨酸消旋酶可能是一个新的治疗靶点。本文从D-丝氨酸和丝氨酸消旋酶的结构性质,作用机理,功能,调控等方面进行了阐述。

1. 引言

D-丝氨酸首先在真核生物被发现,包括蚯蚓,蚕以及其他鳞翅目昆虫中。随着气相色谱和质谱分析应用于手性氨基酸检测和分离,人们发现在哺乳动物神经系统中也存在区域性的高浓度内源性的D-丝氨酸[1]。D-丝氨酸主要存在于哺乳动物脑内灰质区中的Ⅱ型星形胶质细胞内,这种胶质细胞多位于突触旁边[2,3]。在脑部D-丝氨酸主要分布在前脑区,如海马区,纹状体和前额叶皮层,而在其余外围组织分布得很少[4]。D-丝氨酸不同于其它神经元来源的神经递质,作为一种新的神经胶质细胞递质,在神经胶质和神经元传递过程中起着关键的作用[5,6]

2. 丝氨酸消旋酶的结构及性质

在哺乳动物体内富含D-丝氨酸的中枢神经系统中同时发现了丝氨酸消旋酶(Serine Racemase) (EC 5.1.1.18),该酶的发现为内源性D-丝氨酸的产生提供了很好的解释。丝氨酸消旋酶属于Ⅱ型5'-磷酸吡哆醛酶系,结构类似于细菌的丝/苏氨酸水解酶,在ATP和Mg2+的辅助下,通过外消旋作用将L-丝氨酸转变为D-丝氨酸,同时产生硫,氨和丙酮酸盐[7]。Wolosker等通过数据库分析克隆了人的丝氨酸消旋酶,其cDNA全长1020个核苷,编码340个氨基酸,通过与老鼠丝氨酸消旋酶的序列分析,同源性高达88%。Northern-blot分析表明丝氨酸消旋酶主要分布在前脑,在海马区和纹状体处表达量很高,这与D-丝氨酸以及NMDA受体的分布相一致。染色体定位揭示,丝氨酸消旋酶位于17号染色体长臂的13.3处,包含7个外显子[8]。随后从鼠脑中分离纯化了丝氨酸消旋酶,分子量为37KD。丝氨酸消旋酶是一种保守酶,活性较低,对底物L-丝氨酸有高度选择性。酶的最适pH值为8~9,最适温度为37℃。丝氨酸消旋酶保持酶活需要5'-磷酸吡哆醛(PLP),如果去除了PLP,酶活丧失,若添加PLP又会恢复酶活[9]

以丝氨酸消旋酶晶体结构为依据,建立了它的二聚体模型。同时研究表明它存在一些活性部位:PLP结合位点,二价阳离子结合位点,核苷结合位点及蛋白相互作用位点等[10,11](图1)。

3. 丝氨酸消旋酶的催化机理

丝氨酸消旋酶是二聚体,单体的丝氨酸消旋酶有两个结构域,一个大的结构域和一个小的结构域,大的结构域包含可与PLP结合的区域和参与形成二聚体区域[12]。当丝氨酸消旋酶结合ATP和Mg2+,催化L-丝氨酸转变为D-丝氨酸时,酶的构象发生改变,从松弛状态到紧缩状态。这种结构域的变化使得大小两个结构域重新定位,因此小的结构域可能重新定位为催化位点,因而使得关键位点Ser84更有利与底物的结合[13]

丝氨酸消旋酶的催化反应包括消旋作用和底物氨基酸α,β位点的脱氢过程,然后产生对应底物构型正好相反的氨基酸,反应中产生稳定带负电荷的中间物。消旋反应在L-丝氨酸和PLP之间形成了一个醛亚胺中间物(反应1),接着Lys56提取了α位点的质子(反应2)形成了一个稳定的碳负离子中间物[14]。在消旋反应中,在碳负离子中间物的对面质子化作用形成D-丝氨酸(反应3和4)。目前的结构研究证明了这种模式,质子的提取和再质子化是由Lys56和Ser84来完成的,丝氨酸消旋酶催化反应类似于丙氨酸消旋酶[12,13,15,16]。在去质子的反应中,α,β位点的去质子过程是由底物的β-羟基去质子来完成的(反应5),然后脱水(反应6)接着形成了一种不稳定的氨酰中间物,接着氨酰自发的水解形成丙酮酸盐和NH4+(反应7)(图2)。

4. D-丝氨酸的生物学功能

4.1. D-丝氨酸激活NMDA受体的甘氨酸结合位点

NMDA受体是哺乳动物中枢神经系统内一类重要的兴奋性氨基酸受体。它在突触可塑性,学习和记忆等生理过程,以及癫痫、精神病等病理过程中发挥着重要的作用。该受体上存在Gly和Glu结合位点,它的活化需要Gly和Glu位点的共激活[17]。多个研究小组已经证明由丝氨酸消旋酶产生的D-丝氨酸可作用于NMDA受体上的Gly结合位点,通过与Gly位点的高亲和力结合对NMDA受体的功能进行调控,从而影响其生理或病理过程[2,18-20]。生理过程主要体现在NMDA的传导,突触的可塑性以及脑细胞发育过程中的细胞转移等;病理过程主要表现在当D-丝氨酸不能对NMDA受体起到充分的调节作用时,就会引起NMDA受体功能紊乱,引发一些病理过程,如神经精神疾病,衰老,神经恶化性疾病等[21,22]

4.2. D-丝氨酸参与学习和记忆

神经元突触的活动修饰而产生的可塑性被认为是产生学习和记忆的细胞机制。由于海马的长时程增强(LTP)是依赖于NMDA受体的激活,而D-丝氨酸是NMDA受体的内源性配体,表明星形胶质细胞释放的D-丝氨酸与CA1区锥体细胞的LTP的诱导有关。有报道表明丝氨酸消旋酶对NMDA受体应答和LTP是必须的,研究中使用缺失丝氨酸消旋酶的小鼠表现为NMDA受体活性降低而引起的行为性损伤和一定空间的学习性缺乏[23],给予D-丝氨酸表现出了改善学习记忆功能的作用。另外,D-丝氨酸改善D-氨基酸氧化酶对LTP的抑制作用[24]

4.3. D-丝氨酸在神经及精神性疾病治疗中的作用

D-丝氨酸不仅对神经递质的传统观念提出了挑战,而且作为NMDA受体的内源性配体,为NMDA受体过度兴奋或者功能抑制所导致的神经系统疾病提供了新的研究方向。目前的观点认为,直接补充D-丝氨酸可以改善NMDA受体功能低下所致的神经递质传递紊乱,而对于那些NMDA受体过度兴奋引发兴奋性神经毒性反应的疾病,丝氨酸消旋酶可能是一个新的治疗靶点[25]

5. 丝氨酸消旋酶的调控

5.1. 丝氨酸消旋酶被钙、镁等二价阳离子和ATP激活

钙、镁、锰等二价阳离子和ATP均可独立地激活丝氨酸消旋酶[26]。二价阳离子是通过保持丝氨酸消旋酶的稳定性来提高酶的催化效率的。丝氨酸消旋酶中,参与钙结合的残基涉及到Glu210、Asp216和Ala214,这三个残基在植物和酵母丝氨酸消旋酶中都是保守的。但是二价阳离子对酶的激活作用是有限的,当有EDTA存在时会降低酶的催化活性,而ATP仍能增大酶的催化性能,但不会参与酶构象的改变和动力学反应。当有100 µM ATP时,酶的最大反应速率会增大2.2倍,但是米氏常数不变。另外,ATP在催化过程中不会被水解,细胞内ATP的含量远高于活化丝氨酸消旋酶所需要的量,因此推断ATP可能在体内起其他的作用[27]

5.2. D-丝氨酸消旋酶被谷氨酸受体相互作用蛋白(GRIP)的PDZ结构域激活

GRIP包含4个PDZ结构域,PDZ6与丝氨酸消旋酶C末端结构特异结合,PDZ4和PDZ5与AMPA通道结合[28]。还有两个未知功能的GAP结构域。

但是丝氨酸消旋酶仅与GRIP的PDZ6结构域结合并不能被活化,全长的GRIP或者含有PDZ4- PDZ5-PDZ6-GAP2-PDZ7的结构域才能增加酶活。丝氨酸消旋酶与GRIP结合后,酶的最大反应速率和米氏常数都会发生改变。因此推断GRIP的结合引起了丝氨酸消旋酶构象改变从而被激活[12]。另外,与GRIP结合的丝氨酸消旋酶对钙离子的应答曲线不变,这表明,GRIP的结合与钙离子的调节是两条相对独立的路径。丝氨酸消旋酶,GRIP和AMPA的GluR2亚基形成的三元复合物可能使得丝氨酸消旋酶与钙离子通道接近。PICK1含有一个PDZ结构域,它与PKC以及丝氨酸消旋酶的相互作用有关[29,30]。但是还没有研究说明PICK1结合后对于丝氨酸消旋酶活性的影响,目前只是推测PICK1与PKC的相互作用导致瞬时引起了丝氨酸消旋酶的磷酸化[31]

5.3. D-丝氨酸消旋酶活性的反馈调节

在胶质细胞中丝氨酸消旋酶催化产生D-丝氨酸,作用于NMDA受体,引起钙通道的激活而使钙离子进入突触后神经元,接着NO合成酶激活而释放NO,丝氨酸消旋酶随之被亚硝基化而受到抑制[32]

另外,NMDA受体激活后可以使丝氨酸消旋酶从细胞质中转移到细胞膜上,细胞膜上的PIP2通过竞争性的结合ATP来抑制丝氨酸消旋酶的活性,减少D-丝氨酸的生成。这是一种反馈抑制机理,从而防止NMDA受体过度活化[33]

6. 展望

丝氨酸消旋酶存在于哺乳动物中枢神经系统中,转化L-丝氨酸为D-丝氨酸。D-丝氨酸与甘氨酸共同作用于NMDA受体,引起中枢神经系统中信号的传导。科学家在丝氨酸消旋酶的结构及作用机制研究的基础上,进一步对其抑制剂展开研究。目前,最有好的丝氨酸消旋酶抑制剂是L-erythro-3-hydroxyaspartate,类似于底物L-丝氨酸[34]。总之,D-丝氨酸消旋酶可为临床上治疗NMDA受体过度活化引起的神经及精神疾病提供了一种新的作用靶点,为人类的健康带来福音。
