ABSTRACT

MHC gene is a group of highly polymorphic genes. The structure, function and genetic characteristics of MHC gene have been revealed, in recent years, by more and more researchers. This study reviews the MHC in small mammals’ research, mainly on the domestic and foreign small mammals MHC gene polymorphism, and pays attention to the population genetic evolution and gene structure. The future research is prospected.

主要组织相容性复合体(MHC)基因在小型哺乳动物中的研究进展

摘 要

MHC基因是一组多态性非常高的基因群。近年来,MHC基因的结构、功能、遗传学特性被越来越多的研究人员揭示。本文综述了MHC基因在小型哺乳动物中的研究,主要阐述了国内外小型哺乳动物MHC基因的多态性研究,重点包括其在种群遗传进化、基因结构上的研究进展。并对其研究前景进行展望。

1. 引言

主要组织相容性复合体(major histocompatibility complex, MHC)的多态性在基因组中是为最丰富的基因之一,是紧密连锁的一组基因群 [1] 。MHC基因的高度多态性与其自身的基因突变、遗传选择、基因重组和基因转变等作用有关 [2] [3] 。MHC基因编码的蛋白质主要负责识别病原体 [4] [5] 。目前,在国内和国外有关MHC基因的报道很多,主要集中在DP、DQ、DR等几个多态性较高的基因上,特别是它们的第二外显子上 [1] 。故本文以MHC的外显子2基因为分子标记,检测不同小型哺乳动物的遗传多样性。

在哺乳动物中,MHC基因的功能区域被分带为三类:I类、II类,III类 [6] 。MHC I类分子由α1,α2,α3三部分构成:α1和α2结构共同形成抗原肽结合槽(peptide-binding cleft);α3结构域可结合β2微球蛋白。分子内部在α2和α3结构域各形成一个二硫键。在MHC类II分子中,α1和β1形成抗原肽结合槽,分子α2,β1,β2结构域在在分子内部各有一个二硫键。而MHC III类分子主要包括补体因子及细胞因子等。这三类基因在常染色体上以紧密连锁的形式形成单倍型,表现共显性的遗传,又有超基因之称。三类基因上都有很多的基因位点,并且每个基因位点上都存在着大量的等位基因,这使得主要组织相容性复合体基因成为结构最为复杂的遗传多样性系统。由此,在构建模式动物的过程中,大多数遗传学家以MHC基因的遗传多样性,重点分析不同物种间的遗传学特征、免疫学特性,以及对不同物种间的疾病进行分析研究。

2. 研究内容

2.1. MHC基因在树鼩中的研究

在哺乳动物MHC多态性的研究中,树鼩的MHC基因研究相当较少 [7] 。近年来,研究人员在230个树鼩样本中发现了MHC-DRB基因外显子2片段的双引物系统 [8] [9] ,确定了29对等位基因,等位基因命名为TubeDRB-01到TubeDRB-29 [10] 。基因片段总长171bp,其中TubeDRB-21号等位基因只有168 bp,TubeDRB-27号等位基因为终止密码子。

在树鼩的MHC基因研究中发现,所有的等位基因比为dN/dS1 = 2.041 > 1,表明经过了正选择 [11] - [13] ,其中在DRB1等位基因的抗原结合位点上dN/dS = 4.13 > 1明等位基因DRB1经历过正选择且说明了DRB1基因具有高度多态性,并且是每一个样本个体中都能找到的代表性基因。研究还发现只有在TubeDRB-01的等位基因中,抗原结合位点(ABS)上的dN/dS = 4.13 (Z检验,P < 0.05) [14] ,而在所有等位基因上dN/dS = 2.04 (P < 0.01)。说明该基因具有较高的多样性和多态性。

2.2. MHC基因在欧洲棕背䶄中的研究

欧洲棕背䶄的研究主要与寄生虫学、社会生物学相关 [15] 。欧洲棕背䶄能发展成为一个模型动物就在与它的生殖和种群两个有趣的研究上 [16] 。在研究欧洲棕背䶄MHC基因多态性的研究中,科学家们成功用双引物系统的扩增出36个欧洲棕背䶄样本中的MHC-DRB外显子2基因 [17] 。研究表明,36个个体有26对等位基因的存在,其中2号和9号样本被去除,其他样本命名为Clgl DRB-01到Clgl DRB-28。总体样本的核苷酸变异位点数达到70%。表明其核苷酸具有很高的多样性。研究使用MEGA邻接法对样本中的等位基因构建系统发育树 [18] [19] ,发现26对等位基因被分为三大支,其中Clgl DRB-01到Clgl DRB-15号聚为第一支,Clgl DRB-16、Clgl DRB-17、Clgl DRB-18、Clgl DRB-21号聚为第二支,其他聚为第三支。在第一大支中,抗原结合位点和非抗原结合位点间的dN/dS比值差异不显著,但均小于一。其中抗原结合位点的dN/dS < 1 = 0.12,非抗原结合位点dN/dS < 1 = 0.11,说明两者都受到了很强的负选择压力。在第二大支中,抗原结合位点的非同义替换接近为零,无法计算对应的同义替换比例。在第三大支中,虽然没有显著差异,但抗原结合位点的非同义替换远远大于同义替换;恰恰相反,在非抗原结合位点中非同义替换远远小于同义替换。样本总体的dN/dS < 1,说明在欧洲棕背䶄中,MHC基因的进化存在负选择。但第一大支的dN/dS = 0.01远小于整体水平并低于第二大支20倍,说明第一大支的等位基因受到很强大的负选择压力。

2.3. MHC基因在欧亚河狸中的研究

研究发现在76个海狸MHC基因里有10个独特的等位基因,命名为Cafi-DRB-01到Cafi-DRB10 [20] 。基因片段总长有171 bp,其中有15.2%的核苷酸和26.3%的氨基酸可移位点。在两个群体中,除了DRB基因还检测到一个发散序列,代表最有可能的一个假基因。研究发现,总体样本的dN/dS = 3.478 > 1,其中非同义替换(dN)为8.0,同义替换(dS)为2.3。非同义替换几乎是同义替换的四倍。说明研究明确的检测到欧亚海狸MHC-DRB基因进化的正选择,并且该基因有高度的多样性和多态性。

3. 讨论

研究表明,非同义替换与同义替换的比例(dN/dS)是一个被用来证明某个基因经过何种选择的标准指标 [21] 。其中:dN/dS > 1表明该基因或该基因的某个区域经过了正选择;dN/dS < 1表明该基因或该基因的某个区域经过了负选择,dN/dS = 1表明该基因或该基因的某个区域经过了中性选择。

主要以主要组织相容性复合体(MHC)为基础,探讨了小型哺乳动物的种群进化,其中通过同义替换和非同义替换的比值作为标准,说明各小型哺乳动物MHC-DRB基因的进化程度,研究发现,中缅树鼩和欧亚海狸的dN/dS比值均大于一,故两个物种的MHC-DRB基因在进化过程中存在正选择。而在欧洲棕背䶄中,dN/dS比值小于一,说明在欧洲棕背䶄的MHC-DRB基因在进化过程中存在负选择。

通过分析MHC的遗传变异可以提供物种的遗传多样性水平、种群遗传结构进化历史和种群动态等信息,并可在濒危物种的遗传管理中起到重要的作用 [22] 。MHC多样性水平的降低又可导致种群内个体对突发性的传染性病原体的易感性增高,导致种群抗病性能力单一,生存力下降。

4. 展望

在关于小型哺乳动物的研究中,关于主要组织相容性复合体(MHC)的研究还很欠缺,关于MHC基因的其他部分的基因结构和功能也了解甚少,期望在以后的研究中进一步的扩展关于小型哺乳动物MHC基因的研究,进一步的了解小型哺乳动物的进化历程。通过对MHC分子的分析来进行种群遗传学和保护遗传学研究,可以进一步的对模式动物的抗病性,抗病源性能力以及可以为以后的医学方面做出巨大的贡献。
